Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_025144.4(ALPK1):c.74C>T (p.Ala25Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 25 of the ALPK1 protein (p.Ala25Val). This variant is present in population databases (rs369223684, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with ALPK1-related conditions. This missense change has been observed in at least one individual who was not affected with ALPK1-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 2190319). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
