Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.6406_6408dup (p.Thr2136_Ala2137insThr), citing Ambry Variant Classification Scheme 2023: The c.6409_6411dupACA variant (also known as p.T2137dup), located in coding exon 8 of the ALMS1 gene, results from an in-frame duplication of ACA at nucleotide positions 6409 to 6411. This results in the duplication of an extra threonine residue between codons 2137 and 2138. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
