Pathogenic for Leber congenital amaurosis 2; Retinitis pigmentosa 20 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000329.3(RPE65):c.672_673del (p.Glu224fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RPE65 gene (transcript NM_000329.3) at coding-DNA position 672 through coding-DNA position 673, deleting 2 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 224, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu224Aspfs*9) in the RPE65 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RPE65 are known to be pathogenic (PMID: 9326941, 9501220, 9843205, 18632300). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RPE65-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
