Uncertain significance for Congenital myasthenic syndrome 11; Fetal akinesia deformation sequence 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005055.5(RAPSN):c.1030G>A (p.Glu344Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 344 of the RAPSN protein (p.Glu344Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals with RAPSN-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
