Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012079.6(DGAT1):c.629C>T (p.Ser210Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 210 of the DGAT1 protein (p.Ser210Phe). This variant is present in population databases (rs138248019, gnomAD 0.0009%). This missense change has been observed in individual(s) with clinical features of congenital chronic diarrhea (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
