While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 18 amino acid(s) of the DNAJC19 protein. This variant is present in population databases (rs761342359, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with DNAJC19-related conditions. ClinVar contains an entry for this variant (Variation ID: 2185705). This variant disrupts a region of the DNAJC19 protein in which other variant(s) (p.Ala101Profs*10) have been observed in individuals with DNAJC19-related conditions (PMID: 22797137). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
