NM_001378454.1(ALMS1):c.6455A>G (p.Tyr2152Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y2153C variant (also known as c.6458A>G), located in coding exon 8 of the ALMS1 gene, results from an A to G substitution at nucleotide position 6458. The tyrosine at codon 2153 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
