Pathogenic for Nephronophthisis 16 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_173551.5(ANKS6):c.639G>A (p.Trp213Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ANKS6-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Trp213*) in the ANKS6 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ANKS6 are known to be pathogenic (PMID: 23793029, 25599650).
