Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.1295C>A (p.Ser432Tyr), citing Ambry Variant Classification Scheme 2023: The p.S432Y variant (also known as c.1295C>A), located in coding exon 10 of the FBN2 gene, results from a C to A substitution at nucleotide position 1295. The serine at codon 432 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
