It also causes the loss of the initiator methionine codon of the BRCA1 mRNA, and is expected to result in an absent or disrupted protein product. Subgenic deletions in BRCA1 are known to be pathogenic. Deletions of exons 1-2 in particular have been reported in several patients affected with breast cancer (PMID: 24825132, 23640417, 23479189, 21120943, 207276720). For these reasons, this sequence change has been classified as Pathogenic.
