This variant lies in the SLC13A5 gene (transcript NM_177550.5) at coding-DNA position 1207 through coding-DNA position 1217, duplicating 11 bases; at the protein level this means shifts the reading frame starting at proline residue 407, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Pro407Argfs*12) in the SLC13A5 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SLC13A5 are known to be pathogenic (PMID: 24995870, 26384929). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of epileptic encephalopathy (PMID: 26384929). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 218172). For these reasons, this variant has been classified as Pathogenic.
