NM_001378454.1(ALMS1):c.218A>C (p.Asp73Ala) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D74A variant (also known as c.221A>C), located in coding exon 1 of the ALMS1 gene, results from an A to C substitution at nucleotide position 221. The aspartic acid at codon 74 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
