Pathogenic — the classification assigned by GeneDx to NM_032436.4(CHAMP1):c.1544G>A (p.Trp515Ter), citing GeneDx Variant Classification (06012015). This variant lies in the CHAMP1 gene (transcript NM_032436.4) at coding-DNA position 1544, where G is replaced by A; at the protein level this means converts the codon for tryptophan at residue 515 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The W515X pathogenic variant in the CHAMP1 gene has not been reported previously as apathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss ofnormal protein function through protein truncation. The W515X variant was not observed inapproximately 6,500 individuals of European and African American ancestry in the NHLBI ExomeSequencing Project, indicating it is not a common benign variant in these populations. We interpretW515X as a pathogenic variant.
