Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022367.4(SEMA4A):c.212G>A (p.Gly71Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with aspartic acid at codon 71 of the SEMA4A protein (p.Gly71Asp). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is present in population databases (rs774922987, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with SEMA4A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The aspartic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
