Uncertain significance for Glycine encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000481.4(AMT):c.1198T>C (p.Tyr400His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with histidine at codon 400 of the AMT protein (p.Tyr400His). The tyrosine residue is highly conserved and there is a moderate physicochemical difference between tyrosine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with AMT-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C35").
