Uncertain significance for Tall stature-scoliosis-macrodactyly of the great toes syndrome; Acromesomelic dysplasia 1, Maroteaux type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003995.4(NPR2):c.1112G>A (p.Arg371Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 371 of the NPR2 protein (p.Arg371Gln). This variant is present in population databases (no rsID available, gnomAD 0.006%). This missense change has been observed in individual(s) with acromesomelic dysplasia (PMID: 35368703). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 2172426). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt NPR2 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
