Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001080467.3(MYO5B):c.2701C>T (p.Arg901Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 901 of the MYO5B protein (p.Arg901Trp). This variant is present in population databases (rs530225339, gnomAD 0.05%). This variant has not been reported in the literature in individuals affected with MYO5B-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
