Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017950.4(CCDC40):c.2053A>G (p.Ser685Gly), citing Ambry Variant Classification Scheme 2023: The p.S685G variant (also known as c.2053A>G), located in coding exon 13 of the CCDC40 gene, results from an A to G substitution at nucleotide position 2053. The serine at codon 685 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
