Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.18A>C (p.Gln6His), citing Ambry Variant Classification Scheme 2023: The p.Q6H variant (also known as c.18A>C), located in coding exon 1 of the COL3A1 gene, results from an A to C substitution at nucleotide position 18. The glutamine at codon 6 is replaced by histidine, an amino acid with highly similar properties. This variant has been reported in a language impairment cohort (Chen XS et al. Sci Rep, 2017 Apr;7:46105). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
