Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.128A>T (p.Lys43Ile), citing Ambry Variant Classification Scheme 2023: The p.K43I variant (also known as c.128A>T), located in coding exon 2 of the KIT gene, results from an A to T substitution at nucleotide position 128. The lysine at codon 43 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
