Pathogenic for Hereditary insensitivity to pain with anhidrosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002529.4(NTRK1):c.447_450dup (p.His151fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant is also known as c.446_447insTCTG. This premature translational stop signal has been observed in individual(s) with NTRK1-related conditions (PMID: 30774415). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.His151Serfs*23) in the NTRK1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in NTRK1 are known to be pathogenic (PMID: 10982191).
