Uncertain significance for Severe combined immunodeficiency due to DCLRE1C deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001033855.3(DCLRE1C):c.1636C>G (p.Gln546Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with glutamic acid at codon 546 of the DCLRE1C protein (p.Gln546Glu). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and glutamic acid. This variant is present in population databases (rs183622528, ExAC 0.09%). This variant has not been reported in the literature in individuals affected with DCLRE1C-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
