Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.4864G>C (p.Glu1622Gln), citing Ambry Variant Classification Scheme 2023: The p.E1623Q variant (also known as c.4867G>C), located in coding exon 8 of the ALMS1 gene, results from a G to C substitution at nucleotide position 4867. The glutamic acid at codon 1623 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
