NM_001001557.4(GDF6):c.946C>A (p.Pro316Thr) was classified as Uncertain significance for Klippel-Feil syndrome 1, autosomal dominant; Leber congenital amaurosis 17; Isolated microphthalmia 4; Microphthalmia, isolated, with coloboma 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 316 of the GDF6 protein (p.Pro316Thr). This variant is present in population databases (rs556335226, gnomAD 0.05%). This variant has not been reported in the literature in individuals affected with GDF6-related conditions. ClinVar contains an entry for this variant (Variation ID: 2167387). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
