NM_021175.4(HAMP):c.218G>A (p.Cys73Tyr) was classified as Uncertain significance for Hereditary hemochromatosis by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been published in the literature and is not present in population databases. This sequence change replaces cysteine with tyrosine at codon 73 of the HAMP protein (p.Cys73Tyr). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and tyrosine. In summary, this is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
