Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020754.4(ARHGAP31):c.2839C>T (p.Pro947Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with ARHGAP31-related conditions. This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 947 of the ARHGAP31 protein (p.Pro947Ser). This variant is present in population databases (rs748935487, gnomAD 0.0009%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
