This variant was reported in a patient affected with hepatocellular iron storage, hyperferritinemia, and increased transferrin saturation (PMID: 21396368). Segregation studies have not been reported for this variant. Experimental studies have shown that this missense change causes defective hepcidin binding in vitro and leads to hepcidin resistance (PMID: 21396368).
