NM_001378454.1(ALMS1):c.574G>A (p.Glu192Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E193K variant (also known as c.577G>A), located in coding exon 3 of the ALMS1 gene, results from a G to A substitution at nucleotide position 577. The glutamic acid at codon 193 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
