Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005763.4(AASS):c.102C>G (p.Asn34Lys), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 34 of the AASS protein (p.Asn34Lys). This variant is present in population databases (rs777413121, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with AASS-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
