Uncertain significance for Short-rib thoracic dysplasia 11 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_052844.4(DYNC2I2):c.428A>C (p.Gln143Pro), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with WDR34-related conditions. This variant is present in population databases (rs572436409, gnomAD 0.06%). This sequence change replaces glutamine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 143 of the WDR34 protein (p.Gln143Pro).
