Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000124.4(ERCC6):c.2651A>G (p.Lys884Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 884 of the ERCC6 protein (p.Lys884Arg). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ERCC6-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
