NM_013266.4(CTNNA3):c.2472G>A (p.Met824Ile) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M824I variant (also known as c.2472G>A), located in coding exon 17 of the CTNNA3 gene, results from a G to A substitution at nucleotide position 2472. The methionine at codon 824 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
