Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.521G>A (p.Arg174Lys), citing Ambry Variant Classification Scheme 2023: The p.R174K variant (also known as c.521G>A), located in coding exon 3 of the MSH6 gene, results from a G to A substitution at nucleotide position 521. The arginine at codon 174 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
