This variant lies in the NPC1 gene (transcript NM_000271.5) at coding-DNA position 2134, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 712 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 712 of the NPC1 protein (p.Asp712Asn). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This missense change has been observed in individual(s) with Niemann-Pick type C (PMID: 32138288). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 2161649).
