NM_181426.2(CCDC39):c.1644del (p.Asp548fs) was classified as Pathogenic for Primary ciliary dyskinesia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. While this particular sequence change has not been reported in the literature, truncating mutations in CCDC39 are known to be pathogenic (PMID: 23255504). This sequence change inserts 1 nucleotide in exon 12 of the CCDC39 mRNA (c.1644delT), causing a frameshift at codon 548. This creates a premature translational stop signal (p.Asp548Glufs*10) and is expected to result in an absent or disrupted protein product.
