NM_001999.4(FBN2):c.1030G>T (p.Val344Phe) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V344F variant (also known as c.1030G>T), located in coding exon 8 of the FBN2 gene, results from a G to T substitution at nucleotide position 1030. The valine at codon 344 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
