This is predicted to result in a premature translational stop signal and is expected to result in an absent or disrupted protein product. Loss-of-function variants in PALB2 are known to be pathogenic. An exon 11 deletion has been reported in the literature in an individual affected with breast and ovarian cancer. Loss of heterozygosity of the wild-type allele was observed in both tumors (PMID: 24982446). For these reasons, this variant has been classified as Pathogenic.
