Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001142800.2(EYS):c.1471G>A (p.Glu491Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with lysine at codon 491 of the EYS protein (p.Glu491Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs151187049, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with EYS-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
