Pathogenic for Heterotaxy, visceral, X-linked — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003413.3(ZIC3):c.(?_-1)_(*1_?)del, citing Invitae Variant Classification Sherloc (09022015). This is a large deletion in the ZIC3 gene (transcript NM_003413.3) whose exact breakpoints are not precisely mapped. Submitter rationale: A gross deletion of the genomic region encompassing the full coding sequence of the ZIC3 gene has been identified. Gross deletions in ZIC3 are known to be pathogenic. This particular truncation has been reported in a family in which multiple generations of male family members presented with multiple congenital anomalies consistent with heterotaxy and VACTERL-H syndrome. Female carriers from this family were found to be asymptomatic. (PMID: 21465648). For these reasons, this variant has been classified as Pathogenic.
