While this particular sequence change has not been reported in the literature, truncating sequence changes in NF1 are known to be pathogenic (PMID: 10712197, 23913538). This sequence change deletes 2 nucleotides in exon 24 of the NF1 mRNA (c.3138_3139delAG), causing a frameshift at codon 1047. This creates a premature translational stop signal (p.Asp1047Leufs*12) and is expected to result in an absent or disrupted protein product.
