This variant lies in the DNAH5 gene (transcript NM_001369.3) at coding-DNA position 13729, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 4577 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of DNAH5-related conditions (PMID: 27988889). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Arg4577*) in the DNAH5 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 48 amino acid(s) of the DNAH5 protein.
