This leads to an in-frame deletion, preserving the integrity of the reading frame. This variant has not been reported in individuals with a PALB2-related disease. Exon 7 encodes a WD40-like repeat motif in the PALB2 protein. This motif is required for interactions with BRCA2, POLH and RAD51C proteins (PMID: 24141787, 24485656). Deletion of exon 7 is expected to disrupt this domain and very likely disrupts PALB2 function. In summary, this is a rare in-frame gross deletion that eliminates a portion of an important function domain.
