Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001386125.1(OBSCN):c.15015C>A (p.Tyr5005Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Tyr4048*) in the OBSCN gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in OBSCN are known to be pathogenic (PMID: 34957489). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with OBSCN-related conditions. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
