Uncertain significance for 46 XY differences of sex development; Oligosynaptic infertility — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004959.5(NR5A1):c.1093C>T (p.Arg365Trp), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt NR5A1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This missense change has been observed in individual(s) with primary ovarian insufficiency (PMID: 30406445, 34095689). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 365 of the NR5A1 protein (p.Arg365Trp).
