Uncertain significance for Generalized epilepsy-paroxysmal dyskinesia syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001161352.2(KCNMA1):c.186C>G (p.His62Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 62 of the KCNMA1 protein (p.His62Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal dominant KCNMA1-related disorders (internal data). ClinVar contains an entry for this variant (Variation ID: 2158998). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
