NM_000051.4(ATM):c.4335T>G (p.Phe1445Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F1445L variant (also known as c.4335T>G), located in coding exon 28 of the ATM gene, results from a T to G substitution at nucleotide position 4335. The phenylalanine at codon 1445 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
