Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.2003G>A (p.Gly668Asp), citing Ambry Variant Classification Scheme 2023: The p.G668D variant (also known as c.2003G>A), located in coding exon 22 of the RTEL1 gene, results from a G to A substitution at nucleotide position 2003. The glycine at codon 668 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
