NM_004100.5(EYA4):c.511G>C (p.Gly171Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G171R variant (also known as c.511G>C), located in coding exon 7 of the EYA4 gene, results from a G to C substitution at nucleotide position 511. The glycine at codon 171 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been reported in a family with hearing loss and segregated with disease in that family (Liu F et al. PLoS One, 2015 May;10:e0126602). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
