Uncertain significance for Mucopolysaccharidosis, MPS-III-C; Retinitis pigmentosa 73 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152419.3(HGSNAT):c.877A>G (p.Ile293Val), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with HGSNAT-related conditions. This variant is present in population databases (rs779347965, gnomAD 0.0009%). This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 293 of the HGSNAT protein (p.Ile293Val). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt HGSNAT protein function.
