Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.998C>A (p.Ala333Glu), citing Ambry Variant Classification Scheme 2023: The p.A333E variant (also known as c.998C>A), located in coding exon 10 of the RB1 gene, results from a C to A substitution at nucleotide position 998. The alanine at codon 333 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
